Abaloparatide is a synthetic peptide drug produced via recombinant DNA technology, classified as a parathyroid hormone-related peptide (PTHrP) analog. Its amino acid sequence exhibits high homology with endogenous PTHrP(1-34); It specifically binds to the parathyroid hormone type 1 receptor (PTH1R) on the surface of osteoblasts, activating the adenylate cyclase – cyclic adenosine monophosphate (cAMP) signaling pathway. This promotes osteoblast proliferation, differentiation, and collagen synthesis, inhibits osteoblast apoptosis, and modulates osteoclast activity. Consequently, bone formation exceeds resorption, leading to increased bone mineral density, improved bone microarchitecture, and enhanced bone strength.
